Research vision
Healthy and diseased phenotypes of cells and organs are largely defined by the protein composition and protein activities. My vision is to unravel the molecular networks of proteins that maintain cellular homeostasis, and those perturbations that lead to disease, including cancer. We apply mass spectrometry to studies of cancer cells and tissues to identify molecular features of proteins and their roles in disease. We focus on mapping and quantification of post-translational modifications that govern cellular signaling networks and protein-ligand interactions, including protein-protein and protein-DNA interactions in chromatin. We recently established mass spectrometry imaging (MSI) as a new tool to visualize the distributions of biomolecules in tissue sections, including spatial mapping of lipids in fresh frozen tissues and tumors, and proteins in formalin-fixed paraffin embedded (FFPE) tissue and tumor blocks from biobanks.
