Nikon Yokogawa Spinning Disk

Introduction:

Inverted spinning-disk confocal system for high frame rate confocal imaging in light-sensitive or live-cell applications. The microscope setup encompasses environmental control with temperature, humidity and CO control.


Main features and applications:

  • Live-cell imaging with stable temperature, humidity and CO control.

  • High-speed spinning disk confocal imaging across multiple excitation lines.

  • Piezo Z-stage enables fast/accurate imaging in the z-dimension.


Specifications:

Microscope body:
Nikon Ti-E inverted microscope with Perfect Focus System (PFS).

Compatible sample holders:

  • Standard ~25 x 75 mm glass slides + chambered glass slides

  • Culture dishes w. diameter: ~30 – 40 mm

Scan mechanism:
Yokogawa CSU-X1 spinning disk; 50 µm pinholes (optimal for 60×/100×).

Light Sources:

  • Lumencor Celesta Quattro 4ch with Despeckler, lines 405 nm, 476 nm, 555 nm and 637 nm with ~800 mW output per color channel (installed September 2025).

  • Nikon Intensilight for standard EPI illumination when using the eyepiece.

  • Diascopic transmission illumination

Objectives:

  • 20× / NA 0.5

  • 40× / NA 0.75

  • 60× / NA 1.4 oil

  • 100× / NA 1.4 oil

Camera:
Andor iXon EMCCD DU-885 (1004 × 1002 px, 8 × 8 µm) (may be replaced with an Andor Zyla sCMOS).

Stage & Environment:

  • Motorized stage and objective for automated multi-point imaging

  • Nikon Perfect Focus System (PFS) for hardware-based autofocus

  • Prior piezo Z-stage for fast/precise imaging in Z

  • OkoLab cage incubator for controlling humidity, temperature and CO levels.

Software:
NIS-Elements (acquisition) with ND acquisition (and JOBs??).
