Leica SP8 Confocal/CARS/STED

The Leica SP8 is a high-end laser‐scanning confocal microscope system (from Leica Microsystems) designed for advanced biological imaging: multi-colour fluorescence, 3D/time-lapse imaging, super-resolution, live-cell work, etc.

Applications:

  1. Confocal microscopy
  2. Stimulated emission depletion microscopy (STED)
  3. Coherent anti-Stokes Raman spectroscopy (CARS)
  

Confocal:

  • Excitation:  White light laser (WLL) for a flexible choice of up to eight simultaneous excitation wavelengths (470-700 nm).
  • Argon laser excitation at 458 nm, 476 nm, 488 nm, 496 nm, and 514 nm.

Detection

  • Filter-free spectral detection system i.e. option for adjustable detection ranges.
  • Two PMT detectors and one hybrid (HyD) detector(with gated detection).

Objectives

  •  40x/1.10 WATER, HC PL APO CS2
  • 63x/1.40 OIL, HCX PLAPO
  • 100x/1.40 OIL, HC PL APO CS2
  • 20x/0.75 DRY, HC PL APO CS2
  • 10x/0.40 DRY.

Other features

  • Galvano scanner with scan speed up to 7 frames/s (512x512 pixels),
  • Resonant scanner (8 kHz) with scan speeds of up to 28 frames/s (512 x 512 pixels),
  • xyz, xzy, xyzt, xzyt, and lambda scan modes (both excitation and emission).

 

STED

  • The STED (STimulated Emission Depletion) systems is used with the above confocal system.
  • Depletion: The system is equipped with a 592 nm continuous wave (CW) STED laser allowing for super-resolution microscopy.
  • Sub-diffraction limited spatial (xy) resolution of down to ~50 nm standard green emitting STED dyes (eg. alexa488, Atto-488, abberior star 488, chromeo488).

 

CARS and Multiphoton

Laser: 1064nm,

Compatible objectives: 20x Air (IMM) or 40x IRAPO water.
