The Flow Cytometry Facility is equipped with two analytical Flow Cytometers and a high-speed Cell Sorter

The Flow Cytometers

Becton Dickinson LSR II Flow Cytometer:

It is equipped with four lasers:

Violet 405nm, blue 488nm, yellow-green 561nm and a red 639nm.

  • The 405nm and 639nm laser are capable of collecting up to two colors.
  • The 561-nm octagon array is capable of collecting up to four colors.
  • The 488nm laser is capable of collecting up to two colors and side scatter

The configuration with fluorochromes are variable, but the equipment is at present:

  • 405nm laser: AmCyan, Qdot 525, DAPI, Hoechst, V450, BrilliantViolet421, Pacific Blue…
  • 488nm laser:  FITC, Alexa  Flour 488, GFP, CFSE, PerCP, PERCP-CY5,5, PE-CY7 …
  • 639nm laser: APC, Alexa Fluor 647, APC-Cy7, APC-H7…
  • 561nm laser: PE, DsRed, PI, PE-Texas Red, PE Alexa Fluor 610, HcRed, PE-Cy7, PE-Cy5,5

LSR II is also equipped with HTS (High Throughput Sampler) and able to sample from a 96- or 384-microtiter tray.

The Cell Sorter

Becton Dickinson FACSAria III Cell Sorter and Analyser:

 

 

It is equipped with four lasers:

Violet 405nm, blue 488nm, yellow-green 561nm and a red 639nm.

  • The 405nm octagon can detect up to six colors
  • The 488nm laser can collect two colors and side scatter
  • The 561nm octagon array is capable of colleting up to five colors
  • The 633nm laser cant collect up to three colors.

The configuration with fluorochromes are variable, but the equipment is at present:

  • 561nm laser: PE-Alexa Fluor 700, PE-Cy5-5, PE-Texas Red, PI, mCherry, PE-Cy7, PE-Cy5, PE
  • 405nm laser: QDot705, Qdot 605, DAPI, Indo-1, Pacific Blue, QDot 800, Qdot 655, AmCyan
  • 488nm laser: PerCP-Cy5-5, FITC, Alexa fluor 488
  • 633nm laser: APC-Cy7, APC-H7, APC, Alexa Fluor 700

The Cytometer is equipped for two-way and four-way sorting, sorting of cells into 6-well, 24 well or 96-well plates, as well as for single cell cloning. Sorting rates of up to 70,000 cells per second are achievable.

Software and Data Analysis Workstations:

  • The LSR II and the FACSVantage/Diva cell-sorter employ PC-based DIVA software.
  • Data analysis workstations (PC and MAC), accessing FACSCORE shared Network server, are available for data processing with Cell Quest Pro software, DIVA-software and FLOWJo-software.
